چکیده

It has previously been reported that azalomycin F could induce permeability changes in Candida albicans causing loss of cell constituents and inhibition of substrate uptake which resulted in the death of the organism1). The present report is concerned with the effect of the antibiotic on bacteria. Methods and Results Azalomycin F inhibited the growth of Bacillus subtilis PCI 219 at concentrations of 3.0 mcg/ml in Stephanson-Whetham me dium and of 3.0--6.0mcg/ml in nutrient broth, whereas it failed to inhibit the growth of Escherichia coli NIHJ even at concentra tions as high as 200 mcg/ml in both media. In the presence of the antibiotic viable cells of B. subtilis were rapidly reduced in number during the incubation indicating the drug action is bactericidal (Table 1). The release of soluble constituents from washed cells by the action of azalomycin F was then examined in the following way2). The organism was grown overnight in nutrient broth. Cells were harvested by centrifugation, washed thoroughly in phy siological saline, and finally suspended in 1% (w/v) saline to give a suspension containing between 2 and 5 mg dry weight Table 1. Activity of azalomycin F against the cell replication of Bacillus subtilis in natural and synthetic media D ose

The effect of azalomycin F on Ca2+ homeostasis in Trichoderma viride and Saccharomyces cerevisiae.

Azalomycin F (AMF), a macrocyclic lactone antibiotic, in concentrations of 10(-5) g/ml (10(-6) - 10(-5) mol/l) was found to stimulate both the 45Ca2+ influx and efflux in intact Trichoderma viride submerged mycelium and in cells of Saccharomyces cerevisiae without having Ca2+ ionophoric properties. AMF also inhibited ATP-dependent Ca2+ uptake in membrane fractions prepared from T. viride submer...

Mechanism of Azalomycin F5a against Methicillin-Resistant Staphylococcus aureus

To investigate the mechanism of azalomycin F5a against methicillin-resistant Staphylococcus aureus (MRSA), the conductivity of MRSA suspension and the adenylate kinase activity of MRSA culture were determined with the intervention of azalomycin F5a, which were significantly increased compared to those of blank controls. This inferred that azalomycin F5a could lead to the leakage of cellular sub...

Microbiological study of copiamycin.

Copiamycin, an antibiotic agent, has shown in vitro activity against Candida albicans, Torulopsis glabrata, and Trichomonas vaginalis. Local administration of copiamycin to mice inoculated intraperitoneally with protozoa reduced the per cent of infection as measured by decreased abscess formation. This antibiotic has had little effect on the glucose oxidation by protozoa. Its action on anaerobi...

Isolation of three main components. F3, F4 and F5, from azalomycin F-complex.

Isolation of azalomycins F3, F4 and Fg from azalomycin F-complex crystals was successfully conducted. F3 was less soluble in methanol than other components and was isolated by fractional crystallization of F-complex from 80~ 100% methanol in water. F4 and F5 were separated by P-cellulose column chromatography, from which F5 and F4 were eluted with 60 % aqueous methanol and 0.01M ammonium acetat...

Evidence for an iterative module in chain elongation on the azalomycin polyketide synthase

The assembly-line synthases that produce bacterial polyketide natural products follow a modular paradigm in which each round of chain extension is catalysed by a different set or module of enzymes. Examples of deviation from this paradigm, in which a module catalyses either multiple extensions or none are of interest from both a mechanistic and an evolutionary viewpoint. We present evidence tha...
